I grew up in Louisville so I'm biased, but it is indeed a great city. I say skip 4th street as others have alluded to. Bardstown Rd. and Frankfort Ave are filled with great restaurants and bars. For example, the Bristol is a local institution. Great for dinner lunch or brunch and whatever you do just make sure you order the Green Chili Wontons. Louisville is an amazing restaurant city. The park system is pretty great. I recommend Cherokee park very highly....another Frank Law Olmstead design. Other things have already been mentioned and I'd reiterate Churchill Downs if you've never been. If you like old architecture a jaunt through Old Louisville, especially St. James Court is worth it. Cave Hill cemetery is the L'ville equivalent of Spring Grove and you can see Colonel Sanders' grave and feed the ducks. In all it depends what you are into but there is indeed plenty to do.
