I've gotta say I'm really kicking :allovrjr: in the shopping department today. I got a $179.99 pair of earrings for $61 (present for my aunt). Then, I shopped for myself (it's my job) and already got myself Dodgeball, a sweater at Gap, and a jacket. Then, I got my mom a moisturizer at Estee Lauder. And I had already gotten my sister a set of DVD's, so the only person left is my dad and a few friends. I'm definitely in the Christmas spirit. :gac:

Good evening, all.

Momma's working some OT tonight, so I just finished giving the babbies a bath, dinner, and reading "Miffy" to them... seven times... I feel so adult-like.

Neighbor-girl just called and asked me if I'd like to go to Shadowbox Cabaret with her, but... Momma's still at work. She gets off at 6:30 and the doors to the theater close at 7:45, so I told her... maybe. :D

My company christmas luncheon is on the afternoon of Dec 23rd. I asked for this day off and was told I would be expected to be at the luncheon, so I decided to work anyway. This means I have to drive by myself to Houston. If I had the day off, I could have gotten a ride.

So, the luncheon is at Byblos, which is a Lebonese restaurant that the big bosses like very much. It's supposed to start at 1pm, but last year it started late. We'll have wine and food ... it'll last about 2 hours. Employees only. Families not invited. Then I'll have to hop into the car and drive 350 miles to my sister's in Houston. If traffic isn't bad, it take about 6 hours drive time.

Then Sunday, I have to drive back. All so I can attend the company luncheon.

Unless I break down and decide to take greyhound. Airfares that day are just too high.

I was expected to be at a Christmas luncheon too. Since I adopted someone else's soap opera that had to be done yesterday, I stayed in the lab and worked on it. It was cool though. One of the girls brought me back lunch and left it for me in a fridge, since I worked over the weekend.

I'd rather get as much as I can done, and start the new year with a clean slate.

The spousal unit and I braved the Kenwood Mall this AM to finish shopping. It wasn't too bad since we got there early. We parked in the middle of the parking lot at the fringe of the cars. When we came back out, cars stretched out as far as the eye can see.

I was in my normal jeans, Mason sweatshirt, sneakers and was woefully underdressed for the Kenwood mall. All that east Cinci $$$.

The Kenwood mall is 1st on Kronos mall list, it smells nice for starters :D not too many strange people in the parking lot (Tri County has a lot of those), easy to get in and out of (Tri County and Mills both are HORRIBLE at this) and it's a good walk :D

Got lots more wrapping to do, but got a good start last night.

Congrats to LF! Are you sure you want to skip a Chem. level? Looking back now, I wish it was a class I paid better attention to. I did fine in it, but I didn't retain it. Now I need it. Constantly looking stuff up and trying to relearn.

Yeah, I think I'm okay skipping a level. I talked to my professor, my lab supervisor, and my advisor and all felt that I could handle the advanced class with no problem. The only thing is that I may be a little behind in the beginning, but they believe I can pick it up with no problem. So do I. The main reason I'm doing this (besides the fact that OSU offers this option if you get an A in the class that I did), is that it will allow me to take, in the next two quarters, the two Chemistry courses that I need to be eligible to take organic chemistry. This way I can start organic in the fall of next year. I need to have those under my belt by the end of my Junior year so that I can take the MCAT.

I ended up being at a disadvantage when it came to Chemistry because I wasn't able to take it in high school due to my illness. Couldn't do Chemistry at home! So I am behind in that respect and this will allow me to catch up.

I thought the exhibit ran longer than it actually did. So one day I went to see it, and poof, it wasn't there! :( I was so mad at myself for not paying attention to the dates.

If I could go anywhere in the world, Antarctica would be it. It's at the top of my list. It's just so expensive ... although there are trips out there for about half the price of this one. OK, I hope I don't offend anyone by this because I've never heard of this group until just now, but the Scissor Sisters are on SNL right now and they could possibly be the most ridiculous and horrible musical act I've ever seen. Actually, there's no maybe about it. I keep thinking this has to be another skit, but apparently it isn't.

I hope Puffy didn't watch the Giants either... I was forced to against my will. It was the football equivalent of the full body dry heave.

I thought the same about the Browns-Bills game. At this stage of the season you just don't expect to see such sloppy play.

I thought the Bengals played a pretty good game for it being in Foxboro. As usual, it was a typical Patriots game, where they can count on their "D" for at least one score.

The wife and I got to be Mr and Mrs Fix-it this weekend. On Friday the water pressure in the house was phenomemal. The wife said the pressure was so hard that it hurt while in the shower. And I thought she was screaming because of that new 5 speed shower attachment. :D

I went under the house into the crawl space and checked out the pressure switch. As I figured, it wasn't working, and the pump was running constantly, which is not a good thing. You can burn the bad boy up. I had to leave for work, so I turned the pump off at the circuit breaker and told her that when she needed water to turn the pump on, but turn it off when not in use.

Saturday I went to Lowes and got a new switch and installed it. We were back in business. But then, when she was starting to do laundry, the cold water wouldn't shut off filling the washer. So she had to open/cose the cold water valve in order to fill the washer so the water wasn't constantly running.

I figured we could make it through the weekend since we couldn't call the Maytag repair man until Monday. I got on Maytag's website where you can get a on-line manual of your particular model. I determined that it was the dual water valve in the back of the machine. The hot water worked fine. it was just the cold that wouldn't shut off. So the wife and I got into the back panel of the machine, and took the valve off to check it out. At that stage we were assuming the high water pressure may have damaged the valve.

The valve has two diaphrams that open and close, and they were filled with rust and debri. We figured that when I put the new pressure valve in, and purged the water lines, that alot of it must have broke loose and clogged the valve. So we cleaned them and put it all back together.

It works! Saved ourselves about $150 for parts and service call!

Yes thanks Chip. I took a nap before so I'm feeling better. I think it was lingering tiredness from my long trip Friday. But it's been a stressful night, as my grandmother had an allergic rxn to a medicine she was taking. Everything seems to be okay now, but it was scary nonetheless.

Can we make it a new board rule that all new posters must pass an IQ test? People should be at LEAST as smart as Chip to qualify? I know that might be setting the bar a little low, but we have to start someplace?

Ok, so I have determined that I am absolutely clueless about HALF of tomorrow's bio exam material. So I'm begging all of you avid science fans, if you know ANYTHING about protein structure and function, oxygen binding properties for hemoglobin, simple and allosteric binding, Michaelis-Menten enzyme kinetics, allosteric anzyme regulation, and chemotrophic energy metabolism, pretty please come up to Cleveland and take the second half of my final for me tomorrow. Thank you in advance.

(Oh, and I still feel like my stomach is stabbing me from the inside with a butcher knife. Ouchie.) :(

Well, basically everything... I have a decent understanding of the Km and Kd, etc... It must be allosteric stuff I'm not understanding. I'm kinda not getting the whole R form and T form thing with the allosteric binding. Well, I get the idea behind it, but it's pretty tough for me to calculate the K'r and stuff like that for that huge Bb and Bbmax equation... And I'm not sure about the hybrid orbitals involved in the iron in the heme molecule of hemoglobin binding with the protein. All I'm understanding from that is where the oxygen binds and that the hemoglobin has 2 alpha and 2 beta binding sites, which each have one heme.

I'm trying to make sense of my notes from class, along with the notes I just took today and yesterday at the review sessions. Unfortunately, the entire second half of the final is not in the book, which makes for difficult understanding and studying. Plus, the fact that it's an open book-open note exam won't help at all for this part.

Well R and T just refer to "relaxed" and "tense"... where R is the form where oxygen is bound and T is the form where oxygen is unbound. I'm sure you know about cooperativity where the binding of one oxygen makes it easier for the other three to subsequently bind.

As for the iron, hemoglobin contains porphyrin rings, each with an iron center. If I recall correctly, when oxygen binds to the iron center, it converts iron from Fe+2 to Fe+3 (less unpaired e-s). And I believe it's in a square pyramidal geometry, as iron is also coordinated to the imidazole ring of a His residue. If I recall the iron is out of the plane in the unbound form and becomes more planar when the oxygen binds. When the iron is unbound, it is coordinated 5 times -- once to the His residue and 4 times to the porphyrin ring. When it is bound it is coordinated 6 times, the coordinations mentioned above as well as to oxygen.

I did a presentation on this 5 years ago and I also am consulting a friend who is much more educated than I am, so we will get through this.:)

Aww, thank you so much, lgj... You're such a sweetheart!! That definitely helped me with the iron/hemoglobin issue. Right now, I'm putting all my notes in order and in separately labelled folders so I can find information quickly and efficiently during the test. Ok me and my friend just agreed... the iron in the heme center is coordinated 4 times to nitrogens in the porphyrin ring, and once to the imidazole of a His residue in its unbound form, making it a d5. When it binds oxygen it makes it more planar (ie relaxed form), and it becomes d6. It goes from Fe+2 to Fe+3 because there are less unbound electrons. T refers to the Fe+2 form whereas R refers to the Fe+3 form.

Well, I need to know how willing you are to make a quick excursion to Cleveland before tomorrow morning... :MandJ: I think I'm ok for now, thank you so much!! I really appreciate your help!! I just hope I don't let you down and end up flunking it... :(

I'm gonna go over a bunch of different simple and allosteric binding problems, along with some bioenergetics to make sure I can calculate everything I'm supposed to be able to... The good news is that for the second half (the impossible half) of the exam, there are seven questions, but we're allowed to choose five to answer. That makes me feel slightly better, except for the paranoid part of me that worries about selecting the WRONG questions to omit.

I have all my notes, printouts, reviews, quizzes, lab handouts and results, and tests in different labelled folders, all color coded of course for easy identification during the test. I also have color coded and labelled tabs throughout my book, marking important topics as well. My friends are mocking me because of this.
